A mutant cell line partially responsive to both IFN-a and IFN-g BJMBR
A recessive mutant cell line, B7, which is partially responsive to both interferon (IFN)- a and IFN-g is described. B7 was FACS sorted from a cellular pool, which was obtained from the parental cell line 2C4, after several rounds of mutagenesis. The partial responsiveness to IFN was observed both in terms of expression of cell surface markers (CD2, class I and II HLAs) and mRNA expression of IFN-stimulated genes (9-27; 6-16; 2'-5' OAS; GBP and HLA-DRa). A genetic cross with the U4 mutant (JAK1-, a member of the Janus family of nonreceptor tyrosine kinase) did not restore full IFN-responsiveness to B7, and JAK1 cDNA transfection into B7 restored the wild phenotype of the cell...

Detection and evaluation of selection signatures in sheep PAB
Paim,Tiago do Prado; Ianella,Patrícia; Paiva,Samuel Rezende; Caetano,Alexandre Rodrigues; Pimentel,Concepta Margaret McManus.
Abstract: The recent development of genome-wide single nucleotide polymorphism (SNP) arrays made it possible to carry out several studies with different species. The selection process can increase or reduce allelic (or genic) frequencies at specific loci in the genome, besides dragging neighboring alleles in the chromosome. This way, genomic regions with increased frequencies of specific alleles are formed, caracterizing selection signatures or selective sweeps. The detection of these signatures is important to characterize genetic resources, as well as to identify genes or regions involved in the control and expression of important production and economic traits. Sheep are an important species for theses studies as they are dispersed worldwide and have...

Genetic diversity among isolates of stemphylium solani from cotton Trop. Plant Pathol.
MEHTA,Y.R..
The fungus Stemphylium solani causes leaf blight of tomato (Lycopersicon esculentum) in Brazil. In recent years, severe epidemics of a new leaf blight of cotton (Gossipium hyrsutum) caused by S. solani occurred in three major cotton-growing Brazilian states (PR, MT and GO). Molecular analysis was performed to assess the genetic diversity among the S. solani isolates from cotton, and to verify their relationship with representative S. solani isolates from tomato. Random amplified polymorphic DNA (RAPD) markers and internal transcribed spacers of ribosomal DNA (rDNA) were used to compare 33 monosporic isolates of S. solani (28 from cotton and five from tomato). An isolate of Alternaria macrospora from cotton was also used for comparison. RAPD analysis showed...

Genetic variability among the wild boars (Sus scrofa scrofa), crossbred animals and pigs using microsatellite markers (STRs) BABT
Silva,Paula Viana Correa da; Lui,Jeffrey Frederico; Band,Guilherme de Oliveira; Regitano,Luciana Correia de Almeida; Grossi,Selma de Fátima; Sollero,Bruna Pena; Nunes,Cleujosí da Silva.
The aim of this work was to study the genetic variability among the wild boars, crossbred animals and pigs using microsatellite markers. Five genetic groups were studied. The fragments of three microsatellites developed for Sus scrofa domestica - IGF1, ACTG2 and TNFB - were amplified through PCR technique to evaluate the expected intra populacion variability (He) and observed (Ho) heterozygosity, and endogamy coefficient (F IS ) within each population and inter population variability F IS , testing relationship among five genetic groups to establish the genetic distance among them. The high level of observed heterozygosity values varied between 0.537 and 0.7871. Generally, F IS was low, suggesting that the endogamy did not exist between the tested animals.

JAK/STAT-deficient cell lines BJMBR
Bonjardim,C.A..
Mutant cell lines B3 and B10, which are unresponsive to both interferon (IFN)-alpha and IFN-gamma, and line B9, which does not respond to IFN-gamma stimulation, are described. The mutants were submitted to fluorescence-activated cell sorting (FACS) from a cellular pool, which was obtained from the parental cell line 2C4 after several rounds of mutagenesis. The unresponsiveness to IFN stimulation was observed both in terms of expression of cell surface markers (CD2, class I and II HLAs) and mRNA expression of IFN-stimulated genes (2'-5' oligoadenylate synthetase (OAS), 9-27, and guanylate binding protein (GBP)). Genetic crossing of B3, B9 and B10 with U3 (STAT1-), gamma2a (JAK2-) and U4 (JAK1-) mutants, respectively, did not restore IFN responsiveness to...

Since Guzera is a dual-purpose breed, both the MOET Nucleus and various partner herds of the dairy breeding program also participate in the Genetic Evaluation Program of the Guzera Breed for Beef Production (PAGRG) of ANCP and GEMAC. Consequently, several sires are ?double proven?, i.e., they are genetically evaluated for dairy and beef traits. This summary reports for the 11th consecutive year the results of genetic evaluations of beef and reproductive traits in different proven sires for milk. Conformation and management traits can help the breeder to obtain a herd with better production and economic efficiency. Several of these traits are being measured in the Guzera breed and this summary presents the evaluations of sires that met the accuracy...

Origin, evolution and genome distribution of microsatellites Genet. Mol. Biol.
Oliveira,Eder Jorge; Pádua,Juliano Gomes; Zucchi,Maria Imaculada; Vencovsky,Roland; Vieira,Maria Lúcia Carneiro.
Microsatellites, or simple sequence repeats (SSRs), have been the most widely applied class of molecular markers used in genetic studies, with applications in many fields of genetics including genetic conservation, population genetics, molecular breeding, and paternity testing. This range of applications is due to the fact that microsatellite markers are co-dominant and multi-allelic, are highly reproducible, have high-resolution and are based on the polymerase chain reaction (PCR). When first introduced, the development of microsatellite markers was expensive but now new and efficient methods of repetitive sequence isolation have been reported, which have led to reduced costs and microsatellite-technology has been increasingly applied to several species,...
